In our study, we developed a steroid hormones analysis with surface-assisted laser desorption/ionization mass spectrometry (SALDI-MS) method. The Au nanoparticles (AuNPs) were used as SALDI matrices. We tested four steroid hormones:cortisone,hydrocortisone, progesterone, and testosterone. The effects of the concentrations of AuNPs, the concentrations of ammonium citrate buffer, pH of ammonium citrate buffer, and laser fluence on the signal intensity were investigated.The optimum conditions werecontained45 nM AuNPs as matrices prepared in 0.096 mM ammonium citrate (pH 5.0). The laser fluence was 20.62 μJ.Under these optimum conditions, the linear range ofprogesterone, testosterone, and hydrocortisone were from 0.02 to 0.10 mM,and the cortisone was from 0.01 to 0.10 mM, with R2 values of 0.966 or better. This methodwas applied to test of steroid hormones in saliva samples. We compared two liquid-liquid extraction systems includingdiethyl ether and ethyl acetate, and ethyl acetate was better than diethyl ether. The front steps and extraction ratio of acid to add quantitative comparison of response rates and found that the recovery rate of 89% or more.
